地塞米松、油酸、乳酸对体外培养脂肪细胞HSL mRNA表达的影响

摘要 取体外培养生长良好脂肪细胞,培养介质中分别添加0、10、20、40、80、160mg/L地塞米松,0、0.5、1.0、1.5、2.0、2.5mmol/L油酸,0、10、20、30、40、50mg/L乳酸,培养24h后提取总RNA,每个处理3个重复,分别采用荧光定量PCR法检测地塞米松、油酸、乳酸对脂肪细胞HSL mRNA丰度的影响。结果表明油酸、乳酸抑制脂肪细胞内HSL mRNA表达,抑制作用随浓度增加而增强;地塞米松促进脂肪细胞内HSL mRNA表达,是剂量依赖性。 A test was designed to investigate whether hexadecadrol,oleic acid and lactic acid affect the expression of HSL mRNA in adipose cells from a newborn Holstein calf.Hexadecadrol,oleic acid and lactic acid were added to the media with 0,10,20,40、80,160 mg/L and 0,0.5,1.0,1.5,2.0,2.5 mmol/L and 0,10,20,30,40,50 mg/L,respectively.Abundance of HSL mRNA in adipose cells was determined by fluorescent quantitative RT-PCR.The results showed that the expression of HSL mRNA was suppressed in adipose cells treated with oleic acid and lactic acid,with dose dependent respectively.the expression of HSL mRNA was promoted by hexadecadrol with dose dependent.
