弹性蛋白酶损伤肺致炎的地鼠血清中一氧化氮和肿瘤坏死因子的变化 被引量:2

摘要 目的 观察由弹性蛋白酶损伤肺致炎的地鼠血清中一氧化氮 (NO)和肿瘤坏死因子 (TNF- α)的动态变化 ,并初步探讨两者在炎症中的作用。方法 建立弹性蛋白酶损伤肺致炎金黄地鼠模型。用组化和免疫组化方法检测气道上皮细胞和肺内单核巨噬细胞一氧化氮合酶 (i NOS)活性 ,测定血清中 NO和 TNF- α的动态变化。结果 弹性蛋白酶损伤肺致炎地鼠气道上皮细胞和肺内单核巨噬细胞 NOS活性明显加强 ,在炎症早期血清中 NO和TNF- α含量明显增高 ,然后维持在较高水平 (P<0 .0 1)。结论  NO和 TNF- Objective To investigate the role of the NO and TNFα in lung elastase injury induced inflammatory response of hamster. Methods Lung elastase injury induced inflammatory hamster model was established. NOS activity in airway epithelial cells and lung monocytes/macrophage, as well as the serum content of NO and TNFα were measured respectively. Results The results indicated that iNOS activity in epithelium and monocytes/macrophages were significantly enhanced. In the early period of inflammation the NO and TNFα content in serum increased obviously and maintained in a high level (P<0001). Conclusions This Study suggested that both NO and TNFα are important mediators participating in the lung elastase injuryinduced inflammatory response. Our data suggested NO might play a regulatory role in the release of TNFα.
