抑癌基因p^(53)提高人胃癌细胞热敏感性的作用 被引量:15

Enhancement effect of the tumor suppressor p^(53) gene on thermosensitivity of human gastric cancer cell lines

摘要 目的 评价抑癌基因 p5 3对人胃癌细胞 (BGC82 3 )热敏感性的影响。 方法  4种不同 p5 3基因状态的人胃癌细胞系始终用于本实验。用流式细胞光度术 (FCM )分析加热 42℃ 2h或 43℃ 3 0min后 ,在 3 7℃下培养 8h或 16h ,4种细胞系细胞周期分布和凋亡的反应。以 43℃ 2 0min两次加热后的种植肿瘤的抑制曲线比较 4种细胞的热敏感性。结果 在 3 7℃正常条件下 ,4种细胞系细胞周期各时相比例都没有明显的差异。加热 42℃ 2h或 43℃ 3 0min后 3 7℃恢复 8或 16h ,p5 3正常的wtp5 3细胞出现强烈的S期阻滞 ,而且 ,恢复 16h该细胞出现明显的预示凋亡的亚G1峰 ;而在同样条件下 ,其他 3种 p5 3异常的细胞系S期比例没有显著的变化 (P >0 .0 5 ) ,且都没有出现亚G1峰。两次加热wtp5 3细胞种植肿瘤生长比其他 3种细胞肿瘤更明显受到抑制 (P <0 .0 5 )。结论 以上结果证实了 p5 3基因促进了加热后肿瘤细胞的S期阻滞和凋亡 ,从而明显地提高肿瘤细胞的热敏感性。 Objective To assess the role of the tumor suppressor p53 gene on thermosensitivity of human gastric cancer cell lines(BGC823). Methods Four human gastric cancer cell lines with different p53 status were used throught out this study. Cell cycle distribution and apoptosis were analyzed by flow cytometry of cell in 8 h or 16 h at 37 ℃ after heat treatment at 42 ℃ for 2 h or 43 ℃ for 30 min. Plating them into the nude mices, camparing relative volume growth curve after heating at 43 ℃ for 20 min twice. Results BGC823 wtp53 cell with normal p53 had strong arrest in S phase following recover in 8 h or 16 h at 37 ℃ after heat treatment at 42 ℃ for 2 h or 43 ℃ for 30 min, and a sub G 1 peak of apoptosis could be observed following recover in 16 h at 37 ℃ after heat treatment at 42 ℃ for 2 h or 43 ℃ for 30 min, while other three cell lines with abnormal p53 had almost no arrest in S phase and no apoptotic response following same condition. Tumor of BGC823 wtp53 cells after heat treatment at 43 ℃ for 20 min twice significantly was inhibited than those of other three cell lines (P< 0.05 ).Conclusion These results suggst that wild type p53 gene promote S phase arrest and apoptosis of tumor cells following heat shock, and therefore enhance intrinsic thermosensitivity of tumor cells.
