饲喂羊肉和鸭肉对大鼠结肠AQP2表达的影响 被引量:1

摘要 目的:为研究饲喂羊肉和鸭肉对大鼠结肠水通道蛋白2(AQP2)mRNA转录及其蛋白表达水平的影响。方法:将90只3周龄雄性SD大鼠随机均分成3组,分别饲喂豆粕饲料(对照组)、羊肉饲料和鸭肉饲料。饲喂14d和28d对大鼠血清抗利尿激素(AVP)、AQP2mRNA、AQP2蛋白表达及Na+-K+-ATP酶活性进行检测。结果:饲喂羊肉使大鼠的Na+-K+-ATP酶活性、AVP水平、结肠AQP2蛋白表达及其基因转录水平都升高,而饲喂鸭肉使上述指标降低。除结肠AQP2蛋白外,羊肉组和鸭肉组相比,差异显著(P<0.05)。3个实验组大鼠的体质量差异显著,显著顺序为鸭肉组>羊肉组>豆粕组。结论:饲喂羊肉和鸭肉对大鼠结肠AQP2蛋白表达及其基因转录水平具有不同影响,饲喂羊肉使其上调,而饲喂鸭肉使其下调。因此,饲喂羊肉使机体向外利水的生理作用受限,而饲喂鸭肉使机体向外利水的生理作用增强。 Objective: To evaluate AQP2 mRNA transcription and protein expression levels in rats fed die supplemented with sheep meat or duck meat. Methods: Totally 90 three-week-old male SD rats were randomly divided into three groups, and fed a diet supplemented with soybean protein (control), sheep meat or duck meat, respectively. On the 14^th and 28^th day, serum antidiuretic vasopressin (AVP), aquaporin-2 (AQP2) mRNA transcription, AQP2 protein expression, and Na^+-K^+-ATP enzyme activity were determined. Results: The rats fed sheep meat had higher levels of AVP, Na^+-K^+-ATP enzyme activity, AQP2 mRNA transcription and protein expression than those fed duck meat. Significant differences in other indices except protein expression level between sheep meat group and duck meat group were observed. In addition, both feeding groups on the 14~ and 28~ day also showed a significant difference in body weight. Conclusion: Sheep meat and duck meat have different effects on AQP2 mRNA transcription and protein expression. Sheep meetfortified diet reveals a positive effect, while the effect of duck meatfortified is negative. Therefore, sheep meat-fortified diet can limit the release of water, whereas duck meatfortified diet can increase the release of water.
