摘要
为研究饲喂羊肉和鸭肉对大鼠肾脏水通道蛋白2(Aquaporin-2,AQP2)mRNA转录及其蛋白表达水平的影响,将90只3周龄雄性SD大鼠随机均分成3组,分别饲喂豆粕饲料(对照组)、羊肉饲料和鸭肉饲料。饲喂14 d和28 d对大鼠血清抗利尿激素(arginine vasopressin,AVP)、AQP2 mRNA、AQP2蛋白表达及Na+-K+-ATP酶活性进行检测。结果表明:饲喂羊肉大鼠的Na+-K+-ATP酶活性、AVP水平、肾脏AQP2蛋白表达及其基因转录水平均升高,而饲喂鸭肉上述指标均降低,且羊肉组和鸭肉组相比,差异显著(P<0.05)。3个试验组大鼠的体重差异显著,从大到小依次为鸭肉组、羊肉组、豆粕组。饲喂14 d,羊肉组的肾体比显著高于鸭肉组和豆粕组,鸭肉组与豆粕组差异不显著;饲喂28 d,羊肉组的肾体比显著高于鸭肉组。上述结果表明:饲喂羊肉和鸭肉对大鼠肾脏AQP2蛋白表达及其基因转录水平具有显著影响,饲喂羊肉使其上调,而饲喂鸭肉使其下调。因此,饲喂羊肉使机体向外利水的生理作用受限,而饲喂鸭肉使机体向外利水的生理作用增强。
This work was performed to evaluate Aquaporin-2(AQP2)mRNA transcription and protein expression level of rat feed with sheep meat and duck meat.90 three-week male SD rats were randomly divided into three groups,and fed with diet containing soybean protein(control),sheep-meat and duck-meat,respectively.On 14 and 28 days,arginine vasopressin(AVP),Aquaporin-2(AQP2)mRNA transcription,AQP2 protein expression,and Na+-K+-ATP enzyme activity were detected.The results showed that rats fed with sheep meat had significantly higher level of Na+-K+-ATP enzyme activity,AQP2 mRNA transcription and protein expression than those fed with duck meat.After feeding for 14 days,the body weight of three different groups were significantly different,the ratio of kidney and body weight was significantly higher in sheep meat group than that in duck meat group and soybean group,while there was no significant difference between duck meat group and soybean group.After feeding for 28 days,we got similar results with 14 days except for the ratio of kidney and body weight,the ratio of kidney and body weight was significantly higher in sheep meat group than that in duck meat group meat,while there was no significant difference between other groups.The results indicated that sheep meat and duck meat had significantly different effects on AQP2 mRNA transcription and protein expression:fed with sheep meat had a positive effect,while fed with duck meat had a negative influence.Hence,the physiological role of making water outside was limited by feeding with sheep meat while that was increased by feeding with duck meat.
