摘要
目的:观察阿米洛利对L甲状腺素引起大鼠心肌肥厚的作用。方法:大鼠ip L甲状腺素造成心肌肥厚模型后,用形态学方法观察不同剂量阿米洛利对肥厚心肌的作用; 用标准微电极技术记录大鼠右心室肥厚乳头状肌动作电位及低剂量阿米洛利对动作电位诸参数的影响。结果:此心肌肥厚模型能造成心肌肥厚的形态学变化及心肌电生理特性变化(尤其是对APD20 ,50,90 的影响),低剂量阿米洛利能改善肥厚心肌的形态学变化,并能抑制肥厚乳头状肌动作电位时程(APD20) 的延长。结论:低剂量的阿米洛利确能抑制L甲状腺素引起的心肌肥厚,改善肥厚心肌的电生理变化。
AIM: To study the effect of amiloride on L thyroxine induced hypertrophic myocytes in rats. METHODS: Myocardial hypertrophy was induced by L thyroxine (1 mg·kg -1 ·d -1 , ip) in rats. The changes of morphology of rat myocytes under microscope was observed. For recording the action potential of rat papillary muscle, microelectrode technique was used. RESULTS: After administration of thyroxine in rats, the morphology and duration of action potential of the papillary muscle (especially to APD 20,50,90 ) changed. At the dose of 0 5 mg·kg -1 ·d -1 , amiloride(po) was shown to decrease the degree of myocardial hypertrophy and prevent APD 20 of hypertrophic papillary from prolongation. CONCLUSION: At the dose of 0 5 mg·kg -1 ·d -1 , amiloride(po) was shown to be effective against myocardial hypertrophy with methods of morphology and electrophysiology.
