摘要
目的观察仓鼠心肌肥厚模型的心肌Chymase及ACE基因表达的变化。方法:取8周龄仓鼠,行腹主动脉部分结扎术,分别于术后4周、8周处死仓鼠,取心称重并测量心肌壁厚度。采用以β-actin作参照的RT-PCR法对Chymase及ACE基因表达进行检测。结果:显示手术组心重/体重比值及心肌壁厚度明显大于假手术组(P<0.01),表明模型构建成功。手术组动物Chymase基因表达水平随手术后时间的延长有明显增加趋势,手术4周及8周后其表达水平分别增加了2.6倍及12.6倍;而ACE基因表达水平手术后无明显变化。上述结果显示:本文所建心肌肥厚模型中其心肌肥厚的变化可能主要与Chymase基因表达增加有关。提示:Chymase与ACE基因相比前者表达在心肌肥厚中起着更重要的作用。
Aim: To investigate the chymase and ACE gene expression in the left ventricle in hamster myocardial hyper- trophic model. Methods:The abdominal aorta of 8 weeks old hamsters were ligated partly. The hamsters were killed after 4 weeks or 8 weeks of the operation. The weight of heart and the thickness of left ventricular myocardium were measured. The expressions of chymase and ACE genes in left ventricle were detected using RT-PCR with 5-βactin gene expression as reference. Results: After ligation of aorta, the ratio of heart weight to body weight and the thichness of left ventricular myocardium increased compared with that of control(P<0. 01 ). The level of chymase gene expression increased remarkably by 2. 6 and 12. 6 times of control after 4 and 8 weeks of operation, respectively, while little change in ACE was found. Conclusion: The change of hypertrophic myocardium in present studies was mainly associaled with the increase in chymase gene expressioni chymase gene may play a more important role than ACE gene does in the process of myocardial hypertrophy.
