三种色谱法分析鲍鱼生殖腺多糖的单糖组成 被引量:9

摘要 为了选择一种准确有效的方法测定鲍鱼生殖腺多糖(AGP-32)的单糖组成。采用薄层色谱法、气相色谱法和高效液相色谱法对AGP-32的单糖组成进行测定。结果表明:高效液相色谱法较其它色谱方法具有分离效果好,并可同时检测中性糖、糖醛酸和氨基糖等特点,并利用该法得到单糖组成的摩尔比为Man∶Rha∶GlcA∶GalA∶Glc∶Gal∶Xyl∶Fuc=0.79∶2.05∶1.6∶0.58∶0.09∶4.91∶0.11∶0.34。说明高效液相色谱法较其它色谱法更加适合于鲍鱼生殖腺多糖这类复杂样品的单糖组成分析。 To find an accurate and effective method to determine the monosaccharide composition of polysaccharide from abalone gonad(AGP-32).Thin layer chromatography,gas chromatography and high performance liquid chromatography were used to determine the monosaccharide composition of AGP-32.Results: Method of high performance liquid chromatography had good resolution,and it was suitable to simultaneous analyses of neutral,acidic and alkaline monosaccharide.The monosaccharide composition of AGP-32 which identified by this method was Man∶Rha: GlcA∶GalA∶Glc∶Gal: Xyl∶Fuc = 0.79: 2.05∶1.6: 0.58∶0.09: 4.91∶0.11∶0.34.Conclusion: High performance liquid chromatography is more suitable than other chromatographic methods to complicated monosaccharide composition analysis of polysaccharide such as abalone gonad polysaccharide.
