摘要
采用柱前衍生一反相高效液相色谱法对3种百合中13种游离氨基酸含量和组成进行了测定。通过正交试验分析,确定百合中游离氨基酸的最佳提取条件为:乙酸浓度为1%,物料比为1:5,微波功率选择中低火,微波提取时间为2.0rain。结果表明,百合中以精氨酸的含量最高,占总游离氨基酸的40%~50%左右,湖南药用百合中精氨酸含量最高,为13.39mg/g,湖南产地百合游离氨基酸含量高于兰州产地百合含量的40%左右。这可为湖南百合的药用价值研究提供一定的参考依据。
The contents and compositions of 13 free amino acids in three varieties of lily were determined by precolumn derivatization and RP-HPLC. Using orthogonal test to determine the optimal conditions for extracting free amino acids from lily, which are as follows: concentration of acetic acid is 1%, the ratio of material to liquid is 1:5, the microwave power is selected as mid-low, the microwave extraction time is 2.0 rain. The results of column derivatization and RP-HPLC showed that the content of arginine was the highest in lily, which occupied 40%N50% of the content of total free amino acids; the content of arginine in Hunan medical lily was the highest with 13.39 mg/g; the contents of free amino acids in Hunan produced lily were higher than those in Lanzhou produced lily by around 40%. These resuhs provided a certain references for research on medical value of Hunan lily.
