摘要
目的 建立裸鼠胶质瘤动物模型。方法 采用瘤细胞悬液接种法和瘤组织块接种法制作裸小鼠皮下移植瘤 ,并研究肿瘤的病理组织学特征。结果 两种方法所建肿瘤模型其成功率均为 10 0 % ,肿瘤生长状况良好 ,裸鼠无明显恶异质变化 ,肿瘤病理形态学检查符合胶质瘤细胞的形态学特征 ,免疫组化证实体外培养的胶质瘤细胞和体内胶质瘤组织胶质纤维酸性蛋白均高表达。结论 该方法所建裸鼠皮下移植瘤模型能作为研究胶质瘤发病机制。
Objective To develop glioma model in nude mice. Methods The glioma xenografts were developed by means of either C6 cell suspension or tumor tissue pieces inoculated sc into Balb/c nude mice, and the pathology of the tumor tissue was studied. Results Both glioma models in nude mice were successfully developed in 100%, the growth of tumor was well, the pathology of tumor was accord with the morphological features of human glioma cell. The expression of glial fibrillary acidic protein(GFAP) protein was positive in cultured C6 cells and glioma tissue of tumor bearing nude mice. Conclusion The glioma xenograft model in nude mice is a reliable animal model for studying the tumorigenesis mechanism and biological characteristics and gene therapy of glioma.
