摘要
目的 了解鼻咽癌肿瘤细胞增殖与凋亡的关系及bcl 2表达在此过程中的作用。方法 6 5例鼻咽癌活检组织及 15例癌旁组织用免疫组化技术检测bcl 2和PCNA ;HE染色中作核分裂像计数 ,并将后两者合并为增殖指数 ;用原位末端标记法 (ISEL法 )显示 44例鼻咽癌及 15例癌旁组织中的细胞凋亡。结果 ①鼻咽癌PCNA、核分裂像数、凋亡细胞数均高于癌旁上皮组织 ,但与组织分型无关 ;增殖指数与凋亡细胞数呈正相关。②bcl 2在鼻咽癌中高表达 ,并与组织分型有关 ,与凋亡细胞数呈负相关。结论 ①鼻咽癌肿瘤细胞具有高水平增殖的同时也具有高水平的凋亡 ,其肿瘤本身是高水平基础上增殖与凋亡失衡的结果。②bcl 2不是调控肿瘤细胞凋亡的唯一因素 。
Objective To determine the correlation between proliferative and apoptotic indices and detect bcl 2 overexpression in nasophryngeal carcinoma. Methods Immunohistologic staining for bcl 2,PCNA and mitotic counts were performed in archival material from 65 nasopharyngeal carcinomas and 15 normal nasopharyngeal epithelia. The PCNA and mitotic counts were combined into the proliferative indices (PIs). In addition, the apoptotic indices (AIs) were assessed by ISEL in 44 nasopharyngeal carcinomas and 15 normal nasopharyngeal epithelia. Results ①The proliferative indices and apoptotic indices of nasopharyngeal carcinomas were both significantly higher than those of nasopharyngeal epithelia, but not related to histologic classification. The high PIs were associated with the high AIs. ②bcl 2 was detected in the most nasopharyngeal carcinomas. bcl 2 overexpression was related to the histological classification, but negatively correlated with apoptotic indices. Conclusion ①The nasopharyngeal carcinoma has not only high cell growth rate, but also high cell death rate. The development of the tumors in the disturbance of the balance between the proliferation and the apoptosis. ②bcl 2 is not the only factor to regulate the apoptosis, but as an inhibitor of PCD,and it plays a role in the carcinogenesis of the nasopharyngeal carcinoma.
