摘要
以白色念珠菌为指示菌,采用平板琼脂块法进行初筛和发酵液纸片法进行复筛,得到抗真菌活性较强的放线菌8株,对其中一株活性放线菌菌株205-10经16S rDNA方法初步鉴定为阿南德氏链霉菌(Streptomycesanandii),并用不同的温度、pH及紫外线处理发酵液以了解其活性代谢产物的稳定性,用不同有机溶剂进行萃取实验了解其极性.结果表明其活性代谢产物在70℃以下抗真菌活性保持不变,在pH 6~8活性不变,但在紫外灯照射下活性稳定性较不理想,活性物质存在于正丁醇和水中,可知其极性较大.
Eight strains of actinomycetes with strong antifungal activity were found by the plate agar method with Candida albicans as the indicative fungus for initial screening and paper disc method for rescreening. Among them, an active actinomycete No. 205-10 was preliminarily identified to be Streptomyces anandii by 16S rDNA analysis. In order to understand the stability of active metabolites, the fermented liquid was treated with different temperatures, pH and UV irradiation time, and extracted with organic solvents. The results indicated that antifungal active metabolites remained unchanged below 70℃ and pH 6 - 8, but the stability of activities decreased under UV light irradiation. The polarity of the active substances was comparatively large because they existed in the phase of n-butanol and water.
