摘要
对2株分离自新疆乌鲁木齐市的鸡源新城疫病毒F基因进行了扩增、序列测定和分析。结果显示,该2株新城疫病毒F基因核苷酸序列长度分别为1 586bp和1 654bp,分別编码520和545个氨基酸;氨基酸裂解位点的序列均为112R-R-Q-K-R-F117,具备了基因Ⅶ型强毒株的特征;同源性分析表明,该2株新城疫病毒分离株与现用疫苗毒株的同源性在81.8%~84.2%之间,与国内外流行的基因Ⅶ型新城疫病毒同源性在87.4%~98.2%之间,与基因Ⅶd亚型新城疫病毒株的同源性较高,为94.4%~98.2%;遗传进化分析表明,该2个毒株与FJ882014-LN、FJ608335-XJ、FJ80805-JL等国内流行的基因Ⅶd亚型新城疫病毒株在一个分支内亲缘关系最近。
The F genes of two Newcastle disease virus (NDV) isolates, isolated from poultry farm in Urumqi were amplified and analyzed. The results showed that the nucleotide sequence lengths of F genes were 1 589 bp and 1 654 bp,respeetively,encoding 528 and 545 amino acids. The sequence of amino acid at the cleavage site of F gene is 112R-R-Q-K-R-F117, which consistent with the characteristics of Newcastle disease virus of gene Ⅶ. Comparison of the two NDV strains with the current vaccine strains showed that the nucleotide homology was 81.8%-84.2%. Comparison of two NDV strains with the NDV strains of gene Ⅶ of epidemic strains from different countries showed that the nucleotide homology was 87.4%-98.2 %, and compared the nucleotide homology was 94.4 %-98.2 % compared. The phylogenetic tree analysis demonstrated that the two NDV isolates and strains of FJ882014-LN, FJ608335-XJ and FJS0805-JL were located in the same cladogram branch. These strains all belong to NDV of the gene Ⅶd.
