摘要
目的:建立同时测定清肺抑火片中5种蒽醌类化合物(芦荟大黄素、大黄酸、大黄素、大黄酚和大黄素甲醚)含量的方法,并比较不同厂家清肺抑火片中5种蒽醌类化合物的含量差异。方法:色谱柱为AgelaVenusilXBP-C18柱,流动相为乙腈-0.1%磷酸溶液(梯度洗脱),检测波长为226nm,流速为1.0mL.min-1,进样量为5μL,柱温为40℃。结果:芦荟大黄素、大黄酸、大黄素、大黄酚和大黄素甲醚进样量分别在0.010~0.200(r=0.9994)、0.040~0.800(r=0.9993)、0.040~0.800(r=0.9992)、0.040~0.800(r=0.9993)、0.020~0.400μg(r=0.9996)范围内与峰面积积分值呈良好的线性关系;平均加样回收率(n=9)分别为102.95%(RSD=2.13%)、106.47%(RSD=1.88%)、106.48%(RSD=1.41%)、101.47%(RSD=1.45%)、100.31%(RSD=1.88%)。结论:该方法准确可靠、重复性好,可为清肺抑火片的质量控制提供参考;不同厂家的清肺抑火片由于制剂工艺、原材料等不同,导致了5种蒽醌类化合物的含量差异较大。
OBJECTIVE: To develop a method for the content determination of 5 anthraquinone derivatives (aloe-emodin, rhein, emodin, chrysophanol and physcion) in Qingfei yihuo tablets, and to compare the content difference of 5 anthraquinone compounds in Qingfei yihuo tablets from different factories. METHODS: The determination was performed on Agela Venusil XBP-C,8 column with acetonitrile-0.1% phosphoric acid as mobile phase (gradient elution) at the flow rate of 1.0 mL. min-1. The detection wavelength was set at 226 nm, and the injection volume was 5μL; the temperature of column was set at 40℃. RESULTS: The linear ranges were 0.010-0.200μg for aloe-emodin (r=0.999 4), 0.040-0.800 μg for rhein (r=0.999 3), 0.040-0.800μg for emodin (r=0.999 2), 0.040-0.800 μg for chrysophanol (r=0.999 3), 0.020-0.400 μg for physcion (r=0.999 6). The average recoveries were 102.95% (RSD=2.13%), 106.47% (RSD=1.88%), 106.48% (RSD=1.41%), 101.47% (RSD=1.45%) and 100.31% (RSD = 1.88% ), respectively (n=9). CONCLUSIONS: The method developed is accurate, reliable and reproducible, and can be used for quality control of Qingfei yihuo tablets. The content of anthraquinone in Qingfei yihuo tablets from different factories was different, which may result from different raw materials and production process.
