汽油机尾气致大鼠慢性肺损伤及对不同类型胶原蛋白表达的影响 被引量:2

Expressions of Collagen Proteins in Lungs of Rats Exposed to Exhaust of Gasoline Engine

摘要 目的检测汽油机尾气对大鼠慢性肺损伤及不同类型胶原蛋白表达的影响。方法将健康SPF级雄性大鼠30只随机分为6组,即对照组和染毒1、2、4、8、12周组,每组5只,将汽油机尾气通入装有实验大鼠的自制染毒容器中染毒,每天染毒2 h,分别于第1、2、4、8、12周观察肺组织病理学变化,并用免疫组织化学技术检测CollagenⅠ、CollagenⅢ、CollagenⅣ胶原蛋白。结果 Collagen I、CollagenⅢ主要分布于支气管周围与血管周围,阳性表达强弱和面积随染毒时间的延长而增加。CollagenⅣ的表达在染毒2周内与正常对照组无明显差异;但染毒4周后其表达因部位不同而异,肺间隔随染毒时间延长而增厚,肺泡结构破坏,肺泡上皮基底膜的阳性表达面积逐渐减小;但支气管与血管周围阳性表达的强度与面积随染毒时间的延长而增加。结论 CollagenⅠ、CollagenⅢ、CollagenⅣ是汽油机尾气造成大鼠慢性肺损伤时表达的主要胶原蛋白。 Objective To know the effects of gasoline engine exposure on the expressions of collagen proteins the lungs of rats. Methods A total of 30 SD rats were randomly divided into six groups, the control and the exposed groups exposed for one week, two weeks, four weeks, eight weeks and 12 weeks respectively, five rats in each group. The rats were exposed to gasoline engine exhaust in the homemade chambers, for 2 hours a day. All experimental rats were sacrificed in the 1st week, the 2nd week, the 4th week, the 8th week and the 12th week, lung tissues were collected for HE and collagen I, collagen 11I, collagen IV were determined by immunohistochemical technique, the results were analyzed with semi-quantitative processing by image analysis technique. Results Collagen I , collagen llI mainly distributed around the peribronchial and perivascular tissues, the positive signal intensity and its area increased with the exposure time extending. Collagen 1V mainly expressed in the basement membrane of rat alveolar epithelial cells and vascular endothelial cells, its positive area and its intensity decreasing with the extending of exposure time. However, around the peribronchial and perivascular, the positive area and intensity of collagen IV increased with the exposure time extending. Conclusion During the process of lung damage induced by gasoline engine exhaust, the expressions of collagen I , collagen III and collagen IV protein can be affected which may be involved in the damage.
