摘要
用链脲佐菌素 (STZ)作用于离体胰岛β细胞 2 4 h,一氧化氮 (NO)、脂质过氧化产物丙二醛(MDA)生成增多 ,胰岛细胞膜流动性下降 ,微粘度增高 ,同时胰岛素分泌功能降低 ,细胞存活率下降。预先给高浓度褪黑素 (10 μmol/ L)处理 2 4 h者 ,可减少 NO、MDA的释放 ,提高超氧化物歧化酶的水平 ,使胰岛细胞的氧化损伤减轻 ,膜微粘度降低 ,膜流动性增加 ,细胞存活率显著提高。但低浓度褪黑素 (10 nmol/ L)无此作用。提示高浓度褪黑素对 STZ所致的胰岛 β细胞损伤有一定的保护作用。
In order to investigate the protective effect of melatonin on the β cells injury induced by streptozotocin(STZ).STZ was injected cells in vitro for 24h.The results showed that STZ was able to significantly increase nitric oxide (NO)and MDA production, reduce accumulated insulin release and insulin release at glucose stimulation ,decrease the membrane fluidity and cell viability of islets.After 24h pretreatment with 10μmol/L of melationin, it reversed the injury of STZ treated β cells by reducing NO and MDA production and enhancing SOD level.Howerver, 10nmol/L of melatonin had no similar effects.Conclusion:It suggest that melatonin may play a protective role in β cell injury induced by STZ.
