四神丸水煎液中补骨脂素和异补骨脂素的大鼠体内药动学研究 被引量:8

摘要 目的建立测定大鼠血清中补骨脂素和异补骨脂素质量浓度的HPLC法,研究四神丸水煎液(补骨脂、肉豆蔻、五味子、吴茱萸和大枣)中补骨脂素和异补骨脂素在大鼠体内的药动学特性。方法 SD大鼠一次性灌胃给予四神丸水煎液,采用HPLC法测定不同时间点血药浓度,应用DAS2.0软件计算药动学参数。结果补骨脂素和异补骨脂素在血清样品中的标准曲线线性范围分别为0.109~21.80μg/mL(r=0.999 8)、0.097~19.40μg/mL(r=0.999 2),回收率满足要求,且日内及日间精密度RSD均低于15%。补骨脂素和异补骨脂素在大鼠体内的主要药动学参数分别为:t1/2为(4.28±0.68)、(4.97±1.25)h;Tmax为(8.25±0.71)、(8.25±0.71)h;Cmax为(7.85±0.78)、(3.53±0.62)mg/L;AUC(0-∞)为(102.10±15.98)、(51.37±8.33)mg.h/L;MRT(0-∞)为(10.65±0.92)、(11.78±1.49)h。结论该方法简便、快速,适用于补骨脂素和异补骨脂素在大鼠体内的血药浓度测定及药动学研究。 AIM To establish an HPLC method for determining the concentrations of psoralen and isopsoralen in Sishenwan Decoction(Psoraleae Fructus,Myristicae Semen,Schisandrae chinensis Fructus,Euodiae Fructus,Jujubae Fructus) in rat serum and to study their pharmacokinetics characteristics.METHODS Serum samples were collected at different intervals after orally being administered with Sishenwan Decoction and were analysed by HPLC.Pharmacokinetic parameters were calculated by DAS 2.0 software.RESULTS The linear ranges of psoralen and isopsoralen were 0.109-21.80 μg/mL(r=0.999 8) and 0.097-19.40 μg/mL(r=0.999 2),respectively.Their average recoveries met the requirements of methodology,and both RSDs(%) of the within-day and between-day precision were all less than 15%.The main pharmacokinetic parameters of psoralen and isopsoralen were as follows: t1/2 were(4.28±0.68) and(4.97±1.25) h,Tmax were(8.25±0.71) and(8.25±0.71) h,Cmax were(7.85±0.78) and(3.53±0.62) mg/L,AUC(0-∞) were(102.10±15.98) and(51.37±8.33) mg·h/L,MRT(0-∞) were(10.65±0.92) and(11.78±1.49) h,respectively.CONCLUSION The HPLC method for determining the concentrations of psoralen and isopsoralen in rat serum is simple and rapid,suitable for pharmacokinetic studies.
