摘要
运用RAPD分子标记技术,从100条引物中筛选出15条多态性引物,对二代油松种子园的120份样品进行遗传多样性分析。结果表明:种群水平具有较高的遗传多样性,共201个扩增条带中152个为多态性条带,多态位点百分率为75.62%;总的Shannon信息指数(I)为0.4352,Nei’s基因多样性指数(H)为0.3821。采用UPGMA法,在遗传距离0.15处的水平上可将供试材料聚为3类。表明该二代种子园产生一定程度的变异,在分子水平上呈现出遗传多态性。
Genetic diversity and genetic differentiation of Second-generation Seed Orchard of Pinus tabulaeformis Carr. were analyzed by using random amplified polymorphic DNA (RAPD) marker. To better understand their genetic diversity and further utilize them for breeding 120 samples were collected and analyzed using 15 screened RAPD priraers. The results showed that the genetic diversity at species level was relatiely high. The results showed that a total of 201 markers from 200 to 1 000 bp were generated and 152 of them(75. 62%)were polymorphic markers. Shannon information index (I) was 0. 4352 and Nei's gene diver sity (H) was 0. 3821. Using unweightedpair group method with arithmetic average (UPGMA) were classified into 3 groups at level 0. 15,result revealed an abundant genetic diversity of Second-generation Seed Orchard of Pinus tabulaeformis Carr.
