摘要
目的:观察脑血管药CBN对脑血流的作用及在毒性剂量下对脑血流的影响。方法:利用激光Doppler技术测试小鼠大脑皮质运动区脑膜血流量。比较给药前后脑血流的变化。同时监测心电和呼吸。结果:CBN剂量为12.5,25,50mg·kg-1时,有明显增加脑血流的作用,并呈量效关系。大剂量药后10min脑血流明显增加。剂量为800mg·kg-1时脑血流明显下降,其下降50%的平均时间为(11.56±7.54)min,呼吸频率下降25%平均时间为(3.89±0.99)min,心率下降100次/分的时间为(2.4±1.56)min。脑血流下降与呼吸和心率下降的相关系数(r)分别为0.9393和0.9399。结论:药效剂量下CBN有明显增加脑血流的作用。中毒剂量下脑血流下降。这种下降与呼吸及心率的下降呈明显的相关性。提示,CBN急性中毒时主要作用在呼吸及循环系统,并涉及中枢神经系统。
OBJECTIVE:CBN is consisted of notoginsennoside and pueraria flavanoids.To investigate both the effective the dosage and the toxic dosage of CBN on the cerebral blood flow in mice.METHODS:The blood flow on the Rolandos zone—motor area of cerebral cortex (AP -1 ,L 2) was tested by the way of laser Doppler instrument.RESULTS:CBN effectively improved the blood flow,by the dosages of 12.5,25,50 mg·kg -1 ,respectively,with the dose effect relation ship.While the dosage increased to 800 mg·kg -1 (the bottom line of LD 50 ),it decreased obviously the flow.The average time of the flow 50% downwards was at (11.56±7.54) min after CBN injection,the time of the flow 25% decreased respiratory was at (3.89±0.99) min,and the time of the heart rate decreasing 100 times per minute was at (2.4±1.56) min.The correlation coefficients between the blood flow downwards and the respiratory or heart rate decreased were 0.9393 and 0.9399 respectively.CONCLUSION:CBN at low dosage could increase the blood flow.While using the toxic dosage,it could decrease the flow with the obviously correlation between the respiratory and the heart rate.It suggested that the acute toxicity of CBN may be related to the systems of respiratory,circulation and central nervous.
