摘要
对采用海藻酸钠和聚乙烯醇(PVA) 混合的双载体包埋中国红豆杉悬浮细胞的条件进行了初步研究,建立了一套合适的固定化细胞条件:海藻酸钠浓度为30 g/L,聚乙烯醇(PVA) 浓度为70 g/L,CaCl2 浓度20 g/L,包埋密度为0 .18 g/L,接种密度为0 .12 g/L.在此基础上,对中国红豆杉细胞固定化培养的培养基进行了优化,实验结果表明,从红豆杉树皮中分离出的真菌培养物作为诱导物能限制细胞生长,且明显促进紫杉醇合成,并诱导紫杉醇分泌到培养液中.在培养基中适量地添加前体亦能提高紫杉醇的产量.
By studying the conditions of entrapping Taxus chinesis suspense cells with sodium alginate, the suitable immobilizing operation conditions have been established as that the concentration of sodium alginate is 30 g/L, the concentration of polyvinyl alcohol is 70 g/L, the concentration fo calcium chloride is 20 g/L, the trapped cells density is 0.18 kg/L and the inoculations are 0.12 kg/L. On the basis of the above study, the media for immobilized cells of Taxus chiensis are optimized. The experimental results show that the elicitor of the fungi which is screend from Taxus chinensis bark can restrict the cell growth, improve significantly taxol yield and promote excretion of taxol into the medium, and that proper addition of the precursors can also promote the synthesis of taxol.
