摘要
目的:探讨自动化DiversiLab System DNA指纹分型系统(DiversiLab系统)在白假丝酵母同源性分析中的应用,并评价该系统的作用。方法:应用标准化试剂盒进行白假丝酵母的PCR扩增,在微流控芯片电泳和Agilent 2100生物分析仪上收集扩增产物,并用DiversiLab系统分析软件进行DNA指纹分析。结果:35株从重症加强治疗病房患者中分离的白假丝酵母分为7个型别,其中A型13株,B型8株,C型6株,D型3株,E型2株,F型2株,G型1株。2株氟康唑耐药菌株属于B型,4株氟康唑剂量依赖敏感菌株为C型。结论:自动化DiversiLab系统有快速、简便、重复性和分辨率高的特点,且具建立数据库并行不同实验室间数据比对的优势,可作为临床同源性分析鉴定和分型的一线工具。
Objective To apply automated DiversiLab System DNA fingerprinting in homologous analysis of Candida albicans and evaluate its performance.Methods PCR amplification was performed using standardized reagent kits and its product was detected by microfluidics-based electrophoresis and Agilent 2100 Bioanalyzer.The DNA fingerprinting data was interpreted by Web-based software.Results Among the 35 strains of Candida albicans isolated,7 types were classified,including A type(13 strains),B type(8 strains),C type(6 strains),D type(3 strains),E type(2 strains),F type(2 strains) and G type(1 strain).Two isolates with fluconaozle-resistance belonged to B type,and four isolates of fluconazole-susceptible-dose-dependent belonged to C type.Conclusions Automated DiversiLab System offers efficient and excellent discriminatory power with high reproducibility and allows the building of database and inter-laboratory comparisons.It could be recommended as a first-line tool in the identification and typing of Candida albicans.
