摘要 目的研究丹酚酸提取物的HPLC图谱与其抗氧化活性的关系,为建立丹参制剂质量标准提供依据。方法提取总酚酸,纯化制得丹酚酸B,并将总酚酸在不同条件下水解得到7个物质基础有显著性差异的水解产物,共构建出9个丹酚酸样品。进行其抗小鼠红细胞自氧化溶血(模型I)、抗小鼠肝匀浆脂质过氧化(模型II)实验,采用PLS回归建立样品HPLC峰面积与抗氧化活性间的关系,考察各成分对活性的贡献大小与作用方式。结果 9个丹酚酸样品对两个氧化应激模型均有抑制作用,丹酚酸B纯品在两个模型下抗氧化活性均弱于含有小分子酚酸化合物样品。在两个模型下,6个已知的丹酚酸化合物中的丹酚酸B、咖啡酸及原儿茶酸对抗氧化均有正贡献,而阿魏酸有促进氧化的作用;丹参素、原儿茶醛在模型II下有促进氧化的作用,其中丹参素与阿魏酸呈现强的正相关关系。结论丹酚酸提取物抗氧化活性是各种成分协同和拮抗作用的结果。为保证丹参水提制剂的药效、安全性及质量的稳定性(一致性),宜对其中的主要活性成分如丹参素、丹酚酸B质量分数的下限和上限及其相互比例均进行限定。 AIM To study the relationship between HPLC fingerprints and anti-oxidation activity of salvianolic acid extracts to provide evidence for establishing quality standard for Salvia miltiorrhiza preparations. METHODS Total salvianolie acid extract (TSAE) was first prepared and then salvianolic B (SalB) was purified from TSAE. Seven salvianolic acids (SA) samples whose material foundations were significantly different were prepared by hy- drolyzing TSAE under different conditions. Anti-hemolysis of mouse~ red cell caused by auto-oxidation (model I) and mouse~ liver homogenate lipid peroxidation (model II) were used to study antioxidant activity of nine SA sam- ples. A partial least square (PLS) multivariate regression model was constructed to show the correlation between the antioxidant activity of the SA samples from the HPLC fingerprints and peaks possibly responsible for this activi- ty. RESULTS Nine salvianolic acid samples could inhibit hemolysis of mouse~ red cell caused by auto-oxidation or liver homogenate lipid peroxidation. The activity of high purity SalB was weaker in both models above than that of other SA samples involving some small molecule salvianolic acids. In the six known SA substances, SalB, caffeic acid and protocatechuic acid had positive contribution to the antioxidant activity while ferulic acid had negative effect in both models. Danshensu and protocatechuic aldehyde showed negative contribution to the antioxidant acti- vity in model II, and content of Danshensu was highly and positively correlative to that of ferulic acid. CONCLUSION The antioxidant activity of SA extracts shows themself by cooperative and antagonistic effect of many components. In order to ensure the safety, effectiveness and stability of Salvia miltiorrhiza preparations, it is necessary to place the minimum and maximum content limitation and their mutual proportion of the key eomponents, such as Danshensu, SalB.
