摘要
在免疫电镜实验中,大麦黄花叶病毒(BaYMV)萧山分离物经其同源和日本分离物(BaYMV-J)、英国 W 株系(BaYMV-W)和联邦德国 So 株系(BaYMV-So)的抗血清修饰反应中,有些病毒粒子破坏而发生不同程度的解体。改变修饰反应条件,包括改变反应的时间、温度、抗血清工作浓度,用 IgG 代替完整抗血清.用提纯病毒液代替病叶浸出液以及在修饰前用戊二醛固定病毒粒子,均不能抑制和削弱对病毒粒子的破坏;而此病毒分离物对其血清学无关的英国 S 株系(BaYMV-S)抗血清则表现为稳定,从而进一步证实了我国 BaYMV 与日本BaYMV、英国 BaYMV-W 和联邦德国 BaYMV-So 之间有着紧密的血清学关系。病毒在抗血清中不稳定现象可作为确定大麦黄花叶病毒组或其它一些组群内部成员的血清学关系的一种有效方法。
Some particles of barley yellow mosaic virus(BaYMV)isolated from Xiaoshan,Zhejiang were disinteggated to varying degrees after decorations with its homologous antiserum(As)or antisera against BaYMV-J isolate,BaYMV W strains and BaYMV So strain on electron microscopy grids.The disintegration of virus particles could not be inhibited or weakencd whatver by changing the conditions for decoration reaction including time,temperature,antiserum working concentration or the uses of IgG to substitute the intact antiserum,the purified preparation instead of leaf-dip preparation of glutaraldehye to fix the virus particles before decoration.However the virus was stable as attachment of anti-serum to BaYMV English S strain(BaYMV-S).The serological Telationship of these viruses were further affirmed by the disintegration,which can be uscd as a method to determine the relations of barley yellow mosaic virus group or virus in other groups.
