摘要
应用免疫吸附电镜技术研究了我国大、小麦上三种由禾谷多粘菌Polymyxagraminis传线状病毒,即大麦黄花叶病毒(BaYMV)、小麦黄花叶病毒(WYMV)和小麦梭条斑花叶病毒(WSSMV)分离物的血清学关系,结果表明这三种病毒具有共同的抗原性,尤其是WYMV与WSSMV,它们的血清学性质几乎完全相同。我国的BaYMV-萧山(XSh)、嘉善(JSh)、上海(ShH)、扬州(YZh)、盐城(YCh)和常熟(ChSh)6个分离物,均能与日本BaYMV标准分离物(BaYMV-J)的抗血清以及欧洲的So、Wiltshire(W)、NM株系抗血清起反应,表明属欧洲所划分的BaYMV血清型Ⅰ。WSSMV的新昌(XCh)、安吉(AJ)、宜兴(YX)、扬州(YZh)和雅安(YA)6个分离物的血清学性质一致,均能与加拿大和美国的WSSMV抗血清以及日本WYMV抗血清发生反应。讨论了抗血清中甘油的存在对免疫吸附电镜检测结果的影响。
The serological relationships among three Polymyxa graminis-transmit-ted filamentous viruses of cereals, eg. barley yellow mosaic virus ( BaY-MV ) , wheat yellow mosaic virus ( WYMV ) and wheat spindle streakmosaic virus ( WSSMV ) were investigated using immunosorbent electron microscopy. The results showed that all these viruses seem to possess some antigenic determinants in common and cross reactions with each other. All of the six isolates of BaYMV from Xiaoshan ( XSh ) , Jiashan( JSh ), Shanghai( ShH ) , Yangzhou ( YZh ), Yancheng ( YCh ) and Changshu ( ChSh ) showed strong reaction with antisere against typical strain from Japan(BaYMV-J), Europe (BaYMV-So, or Wiltshire ( W ) and BaYMV-NM ), indicating that the Chinese BaYMV isolates were closely related to or identical with representatives of serotype I which included BaYMV-J, So or W and NM. The five isolates of WSSMV from Xingcheng ( XCh ) , AnJi ( AJ ) , Yixing( YX ) ,Yangzhou ( YZh ) and Yaan ( YA ) were identical in serological properties and could react with both American and Canadian antisera to WSSMV. The influence of glycerol as a preservative in antisera on immunosorbent electron microscopy was discussed.
