摘要
目的:探讨草苁蓉提取物对大鼠肝星状细胞株(HSC-T6)生长抑制及诱导凋亡的影响。方法:选用大鼠肝星状细胞株(HSC-T6)进行体外培养,用MTT比色法测定不同浓度、不同时间段草苁蓉乙醇提取物对该细胞的抗增殖作用;应用倒置显微镜、HE染色光学显微镜、吖啶橙/溴化乙啶(AO/EB)双染色荧光显微镜及扫描电镜技术观察药物作用后的细胞形态学改变;应用流式细胞仪(fluorescence activated cell sorter,FACS)检测细胞周期及细胞凋亡率的变化。结果:(1)MTT实验表明:草苁蓉乙醇提取能抑制HSC-T6细胞增殖,并表现出浓度、时间依赖性关系。(2)HE染色、AO/EB荧光染色以及扫描电镜观察发现草苁蓉乙醇提取物作用于HSC-T6细胞后呈现出典型的细胞凋亡形态学改变:细胞表面微绒毛消失,细胞体积变小,细胞质浓缩,核染色质边集、细胞核固缩、核碎裂、出泡等现象。(3)流式细胞仪检测,草苁蓉乙醇提取物处理后大鼠肝星状细胞(HSC-T6)被阻滞于G0/G1期,细胞凋亡率上升,与对照组相比有统计学差异,并随药物浓度的升高呈增高趋势。结论:(1)草苁蓉乙醇提取物在体外可明显抑制HSC-T6细胞的增殖。(2)草苁蓉乙醇提取物在体外可诱导HSC-T6细胞凋亡,并使细胞周期阻滞于G0/G1期。
OBJECTIVE To observe the effects of anti proliferation and inducing apoptosis on the hepatic stellate cell-T6 in dueed by ethanol extract from Bosckniakia rossica. METHODS Hepatic stellate cell-T6 (HSC-T6) was cultured in vitro. The inhibitory effect on the proliferation of HSC-T6 by the ethanol extract from Boschniakia rossica was measured by using the t method of MTT assay. The apoptosis of HSC-T6 was detected by inverted microscope, light microscope and fluorescent microscope, and the ultrastructure of HSC-T6 was observed by scanning electronic microscope. Flow cytometry was used to detect the cell cycle distribution and the apoptotie rate of HSC-T6. RESULTS (1) MTT assay revealed that the ethanol extract from Boschniakia rossica could inhibit HSC-T6 proliferation in a dose- and time-dependent manner. (2)The typical morphological changes of the apoptosis of HSC T6 was detected by light microscope, fluorescent microscope and scanning electronic microscope: cell surface mierovilli disappeared, cell size became smaller, cytoplasm concentrated, nuclear chromatin marginationed, nu cleus pyknosis, karyorrhexis,blebbing. (3)Flow cytometry showed that the HSC-T6 dealed with the ethanol extract from Boschniakia rossica was arrested at G0/G1 phases;the apoptotic rate increased significantly compared with the control group, and it increased as the dose of ethanolic extract from Boschniakia rossiea increaing. CONCLUSION (1)The ethanol extract from Boschniakia rossica had a significant anti proliferation effect on HSC-T6 in vitro. (2)The ethanolic extract from Boschniakia rossica could induce HSC T6 apoptosis and arrest HSC-T6 at G0/G1 phases.
