7-硝基吲唑对大鼠脑缺血再灌流保护作用的研究 被引量:2

A STUDY ON THE PROTECTIVE EFFECTS OF 7-NITROINDAZOLE ON FOCAL CEREBRAL ISCHEMIA- REPERFUSION INJURY IN RATS

摘要 目的 :探讨神经元型一氧化氮合酶抑制剂 (nNOSI) 7-硝基吲唑 (7-nitroindazole ,7-NI)对脑缺血再灌流保护作用的机理。方法 :选用体重 180~ 2 2 0 g的雌性Wistar大鼠 ,随机分组 ,左颈外动脉管腔内置渔线经颈内动脉送至大脑中动脉内 ,使之闭塞。 1h后拔出栓子 ,再灌流 0 5h到 2h。于再灌流前治疗组给予 7-NI(5 0mg kg) ,对照组和假手术组给予花生油 5ml kg ,根据再灌流不同时相 (0 5h ,1h ,2h)再分 3个亚组。缺血再灌流期间定时测定脑匀浆NOS活性和MDA含量 ,用嗜银Ⅲ染色法作光镜病理检查。结果 :再灌流后不同时相 ,7-NI组NOS活性、MDA含量均明显低于单纯缺血再灌流对照组 (P <0 0 5 ) ;光镜检查发现 7-NI组顶页皮质早期变性的神经元数在各时相也均低于相应的对照组 (P <0 0 5 )。结论 :于大鼠大脑中动脉闭塞 1h给予 7-NI(5 0mg kg)后再灌流 0 5h到 2h ,7-NI有如下影响 :抑制NOS活性 ;降低MDA含量 ;减轻光镜下病理的异常变化。证明 Objective:To investigate whether the selective inhibitor of neuronal nitric oxide synthase,7-nitroindazole(7-NI),plays a protec-tive role in the early stage of transient focal cerebral ischemia-reperfusion (IR).Methods:Female Wistar rats weighing 180~220g were divided into different groups at random.The left middle cerebral artery occlusion was achieved by placing a nylon thread from the left external carotid artery to the middle cerebral artery by passing the internal carotid artery.The thread put at the proximal seg-ment of the anterior cerebral artery for 1 hour follwed by the removal of the thread to allow reperfusion for another 0.5-2 hours.At the beginning of the reperfusion,7-NI (50mg/kg) or just its vehicle (peanut oil,5ml/kg) was given intraperitoneally for differentgroups respectively.The rats were randomly divided into 3 main groups: (vehicle-control group;7-NI-treated group;sham-operated group. According to the different reperfusion time point,the main groups were divided into three subgroups respectively.At the end of the experiment,the activity of NOS, the level of cerebral MDA and the neuropathologial changes under optical microscopy were examined.Results:It was found that the activity of NOS and the level of MDA after ischemia reperfusion in 7-NI groups were significantly lower than that in the peanut oil-control groups at different time point (P<0.05.) The abnormalities of optical microscopic changes were also alleviated in the 7-NI groups.Conclusions:The results indicate that postichemic application of 7-NI (50mg/kg) protects the neurons of focal cerebral IR in rats.The neuroprotective effects of 7-NI are mostly due to the inhibition of NO synthesis and the decrease of MDA.
