锰染毒对大鼠肾上腺嗜铬瘤细胞活性氧生成和miR-133b表达的影响 被引量:3

Effect of Manganese Chloride on Generation of Reactive Oxygen Species and Expression of miR-133b in Pheochro-mocytoma Cells

摘要 目的研究锰对大鼠肾上腺嗜铬瘤细胞(PC12)活性氧(ROS)的生成和miR-133b表达的影响。方法调整PC12细胞密度约为1×106/ml,以0(对照)、300、600μmol/L MnCl2分别染毒PC12细胞72 h,采用DCFH-DA荧光探针检测ROS生成情况;以0(对照)、100、300μmol/L MnCl2分别染毒PC12细胞24 h,采用荧光定量RT-PCR检测miR-133b相对表达水平。结果与对照组比较,300、600μmol/L MnCl2染毒组PC12细胞ROS水平升高(P<0.05);300μmol/L MnCl2染毒组PC12细胞miR-133b表达水平上调,而100μmol/L MnCl2染毒组PC12细胞miR-133b表达水平未见改变(P>0.05)。结论 MnCl2可诱导PC12细胞活性氧生成增加及miR-133b的表达上调。 Objective To investigate the effect of MnC12 on induction of ROS generation and miR-133b expression in PC12 cells. Methods ROS generation in PC12 cells was measured by DCFH-DA fluorescent probe,following the PC12 cells treatment with 0,300 and 600 μmol/L of MnC12 for 72 h. The relative level of miR-133b expression was measured by real time RT-PCR,following the PC12 cells treatment with 0,100 and 300 μmol/L MnC12 for 24 h. Results ROS generation in the group treated with 300 and 600 μmol/L of MnC12 were higher than compared with the vehicle control group (P〈0.05). The relative level of miR-133h expression of PC12 cells in 300 μmol/L of MnCl2 group was higher than that of the vehicle control group (P〈O.05). But there was no change in the relative level of miR-133b expression of PC12 cells treated with 100 μmol/L of MnC12. Conclusion MnC12 may induce ROS generation and up-regulate the expression of miR-133b in PC 12 cells.
