岩藻糖在正常人肝细胞、低转移人肝癌细胞和高转移人肝癌细胞表面的含量变化 被引量:3

摘要 目的比较正常人肝细胞、低转移人肝癌细胞及高转移人肝癌细胞表面岩藻糖的含量差异。方法利用能与岩藻糖特异性结合的异硫氰酸荧光素标记的荆豆凝集素(FITC-UEA),与正常人肝细胞、低转移人肝癌细胞及高转移人肝癌细胞悬液充分结合,而后用流式细胞仪定量检测细胞表面的荧光强度,据此评定正常人肝细胞、低转移人肝癌细胞和高转移人肝癌细胞表面岩藻糖的含量。结果正常人肝细胞表面的荧光强度低于低转移人肝癌细胞和高转移人肝癌细胞(P<0.05),高转移人肝癌细胞表面的荧光强度高于低转移人肝癌细胞(P<0.05)。结论细胞表面的岩藻糖含量变化与细胞癌变及癌变细胞的转移潜能有关。细胞癌变后,细胞表面的岩藻糖含量增加,并且癌变细胞的转移潜能越高,其表面岩藻糖含量也越高。 Objective To compare the differential expression level of fucose on the surface of normal hepatocytes,low metastatic hepatocellular carcinoma cells and high metastasis hepatocellular carcinoma cells.Methods The amounts of fucose expressed on normal hepatic cells,low metastatic hepatocellular carcinoma cells and high metastasis hepatocellular carcinoma cells were detected by flow cytometry using fucose-specific fluorescein isothiocyanate-ulex europaeus agglutinin(FITC-UEA) in single cell suspensions.Results The fluorescence intensity detected in hepatocytes was lower than those of low metastatic hepatocellular carcinoma cells and high metastasis hepatocellular carcinoma cells(P0.05 for both comparisons).Compare with low metastatic hepatocellular carcinoma cells,the fluorescence intensity of high metastasis hepatocellular carcinoma cells was greater(P0.05).Conclusion The amounts of fucose on cell surface was found to be closely related to the development and metastasis of hepatocellular carcinoma cells.After cell canceration,the fucose content on cell surface increases,and the higher the metastatic potential of cancerous cells,the higher the content of fucose.
