摘要
目的研究硼卡钠(BSH)对辐射损伤小鼠的抗氧化作用,探讨硼卡钠辐射防护作用的抗氧化机制。方法将BALB/c小鼠按体重随机分为正常对照组、照射对照组、阳性药组(WR2721,200 mg/kg)、BSH低剂量组(20mg/kg)、BSH中剂量组(40 mg/kg)、BSH高剂量组(80 mg/kg),每组7只。各组动物腹腔注射,给药体积均为0.2 ml/20g;正常对照组和照射对照组给予生理盐水;阳性药组照射前连续给药2d,每天1次,其余各组照射前24h给药1次。用60Coγ射线对小鼠进行一次性全身照射,照射剂量为6 Gy,剂量率为0.8 Gy/min。照射后第14天采集小鼠外周血并分离血清,分别检测血清SOD活性和MDA含量。结果 BSH低、中、高剂量组小鼠外周血血清SOD活性均显著高于照射对照组(P<0.05);血清MDA值,低剂量组显著低于照射对照组(P<0.05),中、高剂量组非常显著低于照射对照组(P<0.01)。结论 BSH具有显著的抗氧化作用,提示BSH的抗氧化能力是其抗辐射作用的机理之一。
Objective To investigate the anti-oxygen effect of sodium borocaptate on radiation mice.Methods 42 BALB/c mice was randomly divided into negative control group,model control group,positive control group(WR2721,200 mg/kg),20 mg/kg BSH group,40 mg/kg BSH group,and 80 mg/kg BSH group.The preparations were administrated in peritoneal injection with 0.2 ml / 20g.Negative control group and model control group were administrated with normal saline.Positive control group was administrated twice before irradiation.Irradiated mice were irradiated by γ-ray of 60Co(0.8Gy/ min) with the dose of 6 Gy.In 14 days after being irradiated,SOD and MDA were detected.Results Comparing with model control group,SOD in peripheral blood was increased significantly in the 20 mg/kg.d BSH group,40 mg/kg.d BSH group and 80 mg/kg.d BSH group(P0.05).Comparing with the model control group,MDA was decreased significantly in the BSH treated groups(the 20 mg/kg.d group P0.05,the 40 mg/kg group and the 80 mg/kg group P0.01).Conclusion BSH has the effects of increasing the activity of SOD and decreasing the concentration of MDA on mice.The results suggest that BSH has the anti-oxygen activity and it should be one of the mechanisms of BSH on radioprotection.
