摘要
当归多糖经离子交换柱层析得到3个多糖组分:WASP1由葡萄糖及少量阿拉伯糖和半乳糖组成,为中性多糖;WASP2主要由半乳糖、阿拉伯糖、葡萄糖及糖醛酸(35.38%)组成,为弱酸性多糖;WASP3主要含半乳糖、阿拉伯糖和鼠李糖,糖醛酸含量最高(58.27%),结合单糖组成分析确定是一种果胶多糖.酸水解结合HPAECPAD色谱测定当归多糖中的糖醛酸种类为半乳糖醛酸.通过苯酚硫酸法绘制半乳糖醛酸的干扰标准曲线,消除GalA干扰得到多糖样品的中性糖含量,结合糖醛酸含量从而真实反映了样品的总糖含量.
<Abstrcat>Polysaccharides of Angelica sinensis were separated and analyzed by ion-exchange chromatography. The results showed that W-ASP1 were mainly composed of glucose and short amount of galactose and arabinose; W-ASP2 were composed of galactose, arabinose, glucose and uronic acid (35.38%); W-ASP3 were composed of galactose, arabinose, rhamnose, and high amount of uronic acid (58.27%). It was confirmed that W-ASP3 was a kind of pectic polysaccharide by monosaccharide composition analysis. Acid hydrolysis combined with HPAEC-PAD analysis showed that Angelica polysaccharides contained GalA. GalA interference measurement was made for determination of neutral polysaccharide, the total polysaccharide content was really reflected.
