c-myc、ras与P53基因蛋白在喉癌中的共同表达

摘要 目的:研究癌基因及抗癌基因在喉癌中的联合表达情况。方法:应用LSAB微波免疫组化方法,检测了cmyc、ras 与P53 在30 例喉癌及癌旁粘膜,4 例正常喉粘膜中的共同表达。结果:①ras,cmyc 和P53 的阳性表达率在癌组织中分别为86-7 %(26/30),96-7% (29/30),63-3% (19/30);癌旁粘膜分别为43-3% (13/30) ,56-7% (17/30),10-0% (3/30) ;正常喉粘膜中分别为25-0% (1/4) ,25-0% (1/4) ,0-0% (0/4) 。癌与癌旁及正常粘膜间差异有显著性(P<0-05) 。②2 种以上癌基因蛋白产物同时表达阳性者喉癌组织中有28 例(93-3 % );癌旁组织中14 例(46-7% );而正常喉粘膜中无1 例共同表达。③喉癌组织中,cmyc + ras+ P53,ras + cmyc,P53 + cmyc 同时表达者分别为56-7% ,30-3% ,6-7 % 。④3 种基因产物的表达与喉癌的分化程度无关( P>0-05)。结论:cmyc,ras 及P53 基因的改变均参与了喉癌的发生、发展过程。 Aim:To study coexpression of protein products of oncogene ras,c myc and P53 in laryngeal carcinoma,adjacent mucosa and normal mucosa.Methods:By using LSAB microwave immunohistochemical technique,we studied coexpression of protein products of these 3 kinds of oncogenes in 30 cases of laryngeal carcinoma and adjacent mucosa,meanwhile in 4 cases of normal mucosa.Results:The positive rates of ras,c myc and P53 expressions in carcinoma were 86.7%,96 7% and 63.3% respectively,and 43.3%,56 7% and 10.0% in mucosa adjacent to carcinoma respectively,while those in normal controls were 25.0%,25 0% and 0.0% respectively( P <0 05).Expressions of two or three oncogenes were found in 93.3% of carcinoma, 46.7% of adjacent mucosa and 0.0% in normal laryngeal mucosa.Coexpression rates of c myc+ras+P53,c myc+ras,c myc+P53 in laryngeal carcinoma were 56.7%,30.3% and 6.7% ,respectively.There was no correlation between expressions of c myc,ras and P53 and histological grade ( P >0.05).Conclusion:The alterations of c myc,ras and P53 genes are involved in calcinogenesis and development of laryngeal carcinoma.They may play a combined role in laryngeal carcinogenesis.
