黄芪注射液预处理对大鼠脊髓缺血再灌注模型脊髓功能和神经细胞凋亡表达的影响 被引量:5

Effect of Astragalus Injection Preconditioning on Spinal Cord Function and Neuronal Apoptosis in the Rat Model of Spinal Cord Ischemia-reperfusion Injury

摘要 目的:研究中药黄芪注射剂预处理对大鼠脊髓缺血再灌注模型神经细胞凋亡表达的影响,探讨中药黄芪注射液对大鼠脊髓缺血再灌注模型神经细胞的作用效应及其作用机理。方法:采用大鼠肾动脉分支点下阻断腹主动脉的方法,建立大鼠脊髓缺血再灌注模型。90只SD大鼠采用单纯随机抽样方法分为假手术组(A组,n=30)、缺血再灌注组(B组,n=30)、黄芪注射液预处理组(C组,n=30)。其中假手术组开腹后只暴露腹主动脉不阻断腹主动脉,然后关闭腹腔;缺血再灌注组阻断腹主动脉30min后,放开腹主动脉进行缺血再灌注;黄芪注射液预处理组开腹前,经大鼠尾静脉注射黄芪注射液(10mL/kg),然后开腹阻断腹主动脉30min后,放开腹主动脉进行缺血再灌注。造模完成后,各组动物分别于缺血再灌注12h、1d、2d、3d、7d后用Tarlor法评价大鼠双后肢神经功能,然后取出L2-4段脊髓进行苏木精-伊红染色观察脊髓组织病理学变化,并用原位末端标记法(TUNEL)检测凋亡细胞,计算凋亡指数。结果:黄芪注射液预处理可以减轻大鼠缺血再灌注模型双后肢神经功能损伤情况(P<0.05)、可以减少脊髓缺血再灌注后脊髓神经细胞中凋亡细胞数量(P<0.05)。 Objective:To investigate the effect of astragalus injection preconditioning on spinal cord function and neuronal apoptosis in the rat model of spinal cord ischemia-reperfusion injury,and to explore its relative mechanisms.Methods:The rat model of spinal cord ischemia-reperfusion injury was established by blocking abdominal aorta and then released reperfusion 30 minutes after blocking.All 90 SD rats were randomly divided into sham group(A group,n=30),ischemia-reperfusion group(B group,n=30),and astragalus injection preconditioning group(C group,n=30).The rats in A group exposed but without blocking the abdominal aorta after laparotomy.The abdominal aorta of rats in B group was blocked for 30 minutes,and then released reperfusion.The rats in C group were preconditioned with astragalus injection at the dose of 10 ml/kg 30 minutes before abdominal aorta blocked.The nerve function of the posterior limbs was evaluated by Tarlor methods 12h,1d,2d,3d,and 7d after ischemia-reperfusion injury.The pathological changes of L2-4 spinal cord were observed by HE staining.While,the apoptotic cells were detected by the situ end labeling(TUNEL) method,then calculated the apoptotic index.Results:The neural function scores in C group were obviously higher than B group at different time points(P0.05).The pathological change and apoptotic index in A group was statistically significant different from B group(P0.05).Conclusion: The astragalus injection preconditioning can reduce nerve function damage of the posterior limbs,and reduce the number of apoptotic cells of spinal cord.
