摘要
为给板栗功能成分研究提供参考依据,利用响应面分析法优化板栗仁中多酚物质的提取工艺。以板栗仁中多酚物质的得率为考核指标,研究乙醇体积分数、微波功率、微波处理时间和料液比4个单因素对板栗仁中多酚物质提取的影响。在单因素试验基础上,根据Box-Behnken试验设计原理,设计4因素3水平的响应面分析方法,建立二次多项式回归方程的预测模型,所得微波辅助提取多酚工艺的最佳参数为:乙醇体积分数58%,微波处理时间54s,微波功率895W,料液比1∶30。在此条件下板栗仁多酚理论得率为0.35%,实际测得多酚得率为(0.345±0.004)%,与理论预测值基本相符。
In order to provide the reference for the Chinese chestnut functional component research,the optimal conditions for Chinese chestnut kernel polyphenols were evaluated by means of response surface methodology.Microwave-assisted extraction(MAE) was applied to extract polyphenols from Chinese chestnut kernel.The effects of four independents variables in terms of ethanol concentration,microwave power,extract time,and solid-liquid ratio on the polyphenols yield were determined.On the basis of single-factor experiments,a quadratic polynomial regression equation of the forecasting model was set up by using Box-Behnken design.The optimal extraction parameters to obtain the highest polyphenols yield were ethanol concentration of 58%,time duration of 54 s,microwave power of 895 W,and a solid-liquid ratio of 1∶30(g/mL).The average experimental polyphenols yield under the optimum conditions was(0.345±0.004)%,which agree with the predicted value of 0.35%.MAE method was employed successfully to extract polyphenols from Chinese chestnut kernel.
