宝藿苷-I联合5-FU对食管癌Eca-109细胞增殖及凋亡的影响 被引量:1

摘要 目的:研究宝霍苷-I与5-FU联合应用对食管癌Eca-109细胞增殖和凋亡的影响。方法:实验设联合用药组(12.5mg/L宝霍苷-I+12.5mg/L5-FU)、阴性对照组(0.1%DMSO)、25mg/L宝藿苷-I组和12.5mg/L5-FU组,共4组,每组均设3个复孔,每孔100μl(含Eca-109细胞数为1×10~4个)。作用24、48、72h时,采用四甲基偶氮唑盐(MTT)法分别检测各组Eca-109细胞的增殖;并在作用48h后,用流式细胞术检测Eca-109细胞凋亡率;用Western blot技术检测Bcl-2、Bax蛋白的表达。结果:宝霍苷-I、5-FU单用及其联合应用在24、48和72h时均可明显抑制Eca-109细胞的增殖,与阴性对照组比较,差异均有统计学意义(P均〈0.05),联合应用的抑制作用显著优于单独应用(P〈0.05)。宝霍苷-I、5-FU单独及其联合作用48h后,均可诱导Eca-109细胞凋亡,均可下调Bcl-2蛋白的表达,上调Bax蛋白的表达,与对照组相比差异均有统计学意义(P均〈0.05),且联合用药组较单独用药组作用明显(P〈0.05)。结论:宝霍苷-I与5-FU联合应用对Eca-109细胞的增殖抑制和凋亡诱导有协同作用,可能与下调Bcl-2蛋白的表达、上调Bax蛋白的表达有关。 OBJECTIVE:To study the effects of baohuoside-Ⅰin combination with 5-FU on the proliferation and apoptosis of human esophageal carcinoma cell Eca-109.METHODS:The cells were divided into 4 groups: combination group(12.5 mg/L baohuoside-Ⅰand 12.5 mg/L 5-FU),negative control group(0.1%DMSO),25 mg/L baohuoside-Ⅰgroup and 12.5 mg/L 5-FU group,with 3 holes in each group and 100μl each hole(1×10~4 cells ).After treatment for 24,48,72 h,the inhibitory effect on proliferation of Eca-109 cells was analyzed by MTT.After treatment for 48 h,apoptosis of Eca-109 cells was measured with flow cytometry;the expression of Bcl-2 protein and Bax protein of Eca-109 cells was measured with Western blot.RESULTS:Baohuoside-Ⅰ,5-FU alone and baohuoside-Ⅰin combination with 5-FU inhibited proliferation of Eca-109 cells(P all0.05),the inhibitory effect of the combination group was superior to either alone(P0.05).After treatment for 48 h,apoptosis of Eca-109 cells was induced significantly,the expression of Bcl-2 protein was decreased and Bax protein was increased significantly than control group (P all0.05).The combination group showed more significant effect than either alone(P0.05).CONCLUSION: Baohuoside-Ⅰin combination with 5-FU exerted a synergistic effect on inhibiting proliferation and promoting apoptosis. This effect associated with decreased expression of Bcl-2 and increased expression of Bax.
