摘要
[目的]研究裂叶牵牛花色、叶色等变化机理。[方法]通过田间调查,利用PCR技术从裂叶牵牛蓝白相间突变株幼叶组织中扩增出目的片段,构建重组质粒pMD18-T/Tpn。[结果]田间调查结果发现,变异株的F3代发生分离,叶色突变并不影响花色的变化;PCR扩增结果表明,以蓝白、纯蓝、大花基因组DNA在330 bp处各有1条特异带,而春光、白雪、平安红、垂蔓都没有特异带出现。[结论]变异株重组质粒序列为裂叶牵牛的转座子序列片段。
[Objective]The research aimed at investigating mechanisms of the changes of Pharbitis.nil flower colors and leaf colors.[Method] The recombinant plasmid pMD18-T/Tpn was constructed by amplifying the target fragments from young leaf tissues of the blue and white mutants of pharbitis nil with PCR technology and investigating in the fields.[Result] The results of investigations in the fields suggested that the mutants were segregated in F3 generation,and the mutations of leaf colors did not affect the changes of flower colors.The results of PCR showed that there was a band about 330 bp in genomic DNA of Lanbai,Chunlan,Dahua,respectively,however,no bands were found in Chunguang,Baixue,Ping’anhong and Chuiman.[Conclusion] The sequence of recombinant plasmid of the mutant was indeed a fragment of the transposon of pharbitis.nil.
