康脑液对脑缺血再灌注损伤大鼠血清及脑组织中氨基酸含量的影响 被引量:8

Effect of KangNaoYe on the Content of Amino Acids of Serum and Brain Tissues in Rat Model of Cerebral Ischemia-Reperfusion

摘要 目的:观察康脑液对SD大鼠脑缺血再灌注损伤后血清及脑组织中氨基酸水平的影响,并探讨其对脑缺血再灌注保护作用的可能机制。方法:采用线栓法复制大鼠大脑中动脉栓塞模型(MCAO),将36只SD大鼠随机分为假手术组,模型组,康脑液低、中、高剂量组,依达拉奉组,每组6只。脑缺血2h再灌注24h后断头取血、取脑,应用高效液相色谱法测定各组大鼠血清及脑组织中天冬氨酸(Asp)、谷氨酸(Glu)、甘氨酸(Gly)和γ-氨基丁酸(GABA)水平。结果:与假手术组相比,模型组大鼠血清及脑组织中Asp、Glu、Gly、GABA水平明显升高(P<0.05);与模型组比较,康脑液能够降低脑缺血再灌注损伤大鼠血清及脑组织Asp、Glu水平,升高Gly、GABA水平,差异均有统计学意义(P<0.05)。结论:康脑液能够减轻大鼠脑缺血再灌注损伤,机制可能与其影响血液及脑组织内兴奋性氨基酸及抑制性氨基酸代谢平衡有关。 Objective: To observe the effect of KangNaoYe on the content of amino acids (AA)in serum and brain tis- sues in rat model of cerebral ischemia-reperfusion, and the mechanism thereof. Methods: Thirty-six rats were randomly di- vided into 6 groups, sham-operat!on group, ischemia-reperfusion group, KangNaoYe group and edaravone group. The models of cerebral ischemia-reperfusion were established in rats by methods of middle cerebral artery occlusion (MCAO). The ani- mals were killed 24 h after perfusion. The serum and brain tissue levels of aspartic acid (Asp), glutamate (Glu), glycine (Gly), aminobutyric acid (GABA) were detected by reversed-phase high performance liquid chromatography with pre-column de- rivatization respectively. Results: Compared with the shamed group, the contents of Asp, Glu, Gly and GABA were increased significantly in model group (P 〈 0.05). The levels of Gly, GABA activities were increased significantly, but Asp and Glu levels were decreased significantly in KangNaoYe group compared with those of ischemia-reperfusion group (P 〈0.05). Conclusion: KangNaoYe showed protective effects on cerebral ischemia-reperfusion injury in rats, which may be by mediating the level of AA in blood serum and brain tissues.
