摘要
为探讨不同含水量植物叶片进行扫描电镜观察的最合适的样品处理方法,及不同电子束加速电压对扫描电镜图像清晰度的影响,对不同含水量叶片进行了比较性研究。并通过理论分析得出:含水量小于75%的植物叶片可不经处理,直接进行观察,大于79%的叶片必须经过样品制备后观察;5-15KV加速电压时获得的图像最清晰。
In order to select the appropriate sample treatment metholds of plant leaves for SEM observation, and to investigate the effect of electron beam accelerating voltage on SEM image resolution, plant leaves with different water contents were observed with SEM and the results were compared. It was showed that the leaves with water contents less than 75% could be observed directly, while leaves with water contents greater than 79% should be observed after treatment. The images obtained at accelerating voltages of 5-15kV had the highest resolution.
