摘要
收集不同的虫草菌株,对5个虫草菌株进行了分离纯化、形态学观察、人工栽培和分子鉴定等试验,研究了正常菌株和退化变异菌株间的差异.并从中筛选出最适合开发利用的优良菌种。结果表明:蛹虫草不同菌株间生物学特性的差别属于种类差异。在人工栽培试验中,K17菌株的子实体长,颜色橘黄,出草整齐,子囊壳丰富,产量高,每瓶干重达4.67g。K17菌株是5个供试菌株中最适宜人工栽培和开发利用的菌种。5个虫草菌株的ITS序列长度为545-566bp,GC含量为56.11%-57.06%。经核酸序列数据库GenBank同源性检索比对,均与蛹虫草有高度同源性,且5个菌株亲缘关系很近。
In this study, different strains of Cordyceps strains were collected. In order to research the difference in normal strainsand mutations and screen superior strains, comparison research was made on five of them in terms of isolation and purification, morphological observation, artificial cultivation and molecular appraisal. The results indicated that difference of biological characteristics among these strains of Cordyceps militaris is the difference was species. In artificial cultivation, the fruiting body of K17, which was long, orange color, grew symmetrically in the container. The tip of fruiting body contained abundant perithecia. Dry weight of each bottle could be as high as 4.67 g. So K17 was the most suitable strain for artificial cultivation and development. ITS sequences were 545-566bp in length with the GC content of 56.11%-57.06%. The tested strains were similar to Cordyceps militaris in terms of homology from GenBank database and their relatives were very close.
