链脲菌素制备糖尿病小鼠模型影响因素的探讨 被引量:3

摘要 目的探讨链脲菌素(STZ)一次性注射制备糖尿病小鼠的影响因素。方法采用不同给药剂量、小鼠体重及给药途径等因素来评价STZ制备糖尿病小鼠情况,并优选出最适的条件。结果当STZ给药剂量为130mg.kg-1时,小鼠的3天死亡率及7天转阴率最低;高体重组(24~28g)小鼠造模后存活率明显高于低体重组(18~22g);静脉给药的造模成功率高于腹腔注射给药。结论采用体重24~28g小鼠,静脉注射STZ 130mg.kg-1能制备较好的糖尿病小鼠模型。 OBJECTIVE To investigate the factors affecting the diabetic model induced once by STZ in Mice.METHODS Evaluating the situation in STZ diabetic mice and optimizing optimal conditions by the factors of different dosage,route of admini-stration and mice body weight.RESULTS When the dose of 130mg·kg-1 STZ admini-stration,the mice 3 days′ rate of mortality and 7 days′ rate of negative were lowest;high-weight group(24~28g) mice were significantly higher than the survival rate after the model of low weight group(18~22g);intravenous administration of modeling success rate higher than intraperitoneal injection.CONCLUSION 24~28g body weight in mice,intravenous injection of STZ 130mg·kg-1 to prepare better mouse model of diabetes.
