摘要 [目的]建立猪嗜血支原体的PCR诊断方法,并与常规染色镜检诊断方法进行比较。[方法]对采自新疆阿拉尔垦区的疑似样品分别采用吖啶橙、姬姆萨染色镜检,采用温度法将嗜血支原体从红细胞上解离下来,提取其基因。根据GenBank上发表的猪嗜血支原体16S rRNA基因组序列(U88565)设计1对特异性引物,进行PCR扩增,建立猪嗜血支原体的PCR诊断方法。[结果]比较了3种诊断方法,指出了各自的优劣。所建立的PCR方法为猪嗜血支原体病的快速诊断及流行病学调查提供了新手段。 [Objective] The aim was to establish PCR diagnose method for Mycoplasma suis and compare with conventional staining method.[Method] The suspected samples from Alar,Xinjiang were stained by acridine orange and giemsa,then the gene of Mycoplasma suis were extracted which were dissociated from the blood cells at the appropriate temperature conditions;according to Mycoplasma suis 16S rRNA(U88565) published in GenBank,a pair of specific primers were designed,PCR-based assay was established.[Result] Three diagnostic methods were compared,their advantages and disadvantages were pointed out.The established PCR method for Mycoplasma suis was a rapid and new diagnosis means for epidemiological investigation of Mycoplasma suis.
