猪嗜血支原体病原染色检测法的比较研究 被引量:4

摘要 为检测猪嗜血支原体(M.suis)感染,本研究对传统血涂片吖啶橙染色方法进行改良,建立了快速检测M.suis的血液吖啶橙染色方法。结果显示,血液吖啶橙染色法的染色时间比传统血涂片吖啶橙染色法和姬姆萨染色法分别节省约30 min和1 h。利用血液吖啶橙染色法、血涂片吖啶橙染色法、姬姆萨染色法及PCR法对200份临床血液样品进行检测,结果显示,阳性检出率依次为64.5%、59.5%、48%及68.5%。3种染色方法与PCR的符合率分别为87%、77%、59.5%;与PCR结果相比,前两种染色方法差异不显著(p>0.05),后者差异极显著(p<0.01)。结果表明,血液吖啶橙染色法优于其他两种染色方法,具有操作简单快速、准确度高、检出率高等优点,适用于临床上对M.suis感染快速定性检测和流行病学调查研究。 In the present study, we established the acridine orange staining blood assay (AOSBA) for the rapidly detecting Mycoplasma suis in clinical blood samples. Results showed that the established assay saved nearly 30 minuses and 1 hour, respectively, compared with acridine orange staining blood smear and Giemsa staining blood smear. The AOSBA, acridine orange staining blood smear, Giemsa staining blood smear and PCR assay were used to detect 200 blood samples which collected from swine-farm, and the positive rates were 64.5%, 59.5%, 48% and 68.5%, respectively. Compared with PCR assay, the coincidence rate of three staining methods were 87%, 77% and 59.5%, respectively. There were no significant difference of the positive rate between the AOSBA and PCR assay (p〉0.05), between acridine orange staining blood smear and PCR assay (p〉0.05). However, the positive rate of Giemsa staining blood smear and PCR assay was significant different (p〈0.01). The study results showed AOSBA was more sensitive than that of other two staining methods, which was suitable for rapid clinical dectection and epidemiologic survey of M.suis infection.
