摘要
采用左心室及血管插管法,研究了洛土辛(Lot)对大鼠及狗血流动力学的影响。 Lot 2.5,5,10 mg/kg,iv可剂量依赖性地增加麻醉大鼠的左心室压(LVP)、左室压力变化最大速率(±dp/dtmax)、动脉收缩压(SAP),降低左室舒张末期压(LVEDP),但对心率(HR)和动脉舒张压(DAP)却无明显的影响。麻醉大鼠 Lot 5 mg/kg、麻醉狗 Lot 3 mg/kg,iv可获相似结果,其对狗血流动力学影响的起效时间较快且作用持续较长。在毁脊髓大鼠,Lot 5 mg/kg,iv不仅显著地增加 LVP,±ap/dtmax,SAP,明显增加DAP和HR。提示Lot对血流动力学影响的特性与PDE Ⅲ抑制剂相似。
Lotusine (Lot ), a pure alkaloid extracted from the green seed embryo of Nelumbo nucifera Gaertn, has been demonstrated to possess the properties of phosphodiesterase Ⅲ inhibitors in isolated myocardium and blood vessels. In this paper, the effects of Lot on haemodynamics were studied in rat and canine, Lot 2.5, 5, 10 mg/kg, iv could obviously increase left ventricular pressure (LVP), systolic or diastolic maximum rate of pressure change (±dp/dt), left ventricular end diastolic pressure (LVEDP) and systolic artery blood pressure (SAP) in dose dependent manners. But there is no significant influence on diastolic artery blood pressure (DAP) and heart rate (HR ). In anesthetized rat, with Lot 5 mg/kg, iv and anesthetized canine with Lot 3 mg/kg, iv, results similar to the above could be obtained. Moreover, it was found that the effect of Lot on anesthetized canine appeared earlier and was more persistent than that on anesthetized rat. In pithed rat, Lot 5 mg/kg, iv enhanced not only LVP, ±dp/dt, SAP, but also DAP and HR. The results suggested that the properties of the effect of Lot on haemodynamics are similar to that of phosphodiesterase Ⅲ inhibitors.
