硅胶柱分离纯化发酵液中叶酸的工艺研究 被引量:1

Study on separation and purification of folic acid from fermented broth by silica gel column chromatography

摘要 采用硅胶柱层析法分离纯化产朊假丝酵母Y2.12发酵液,在考察洗脱剂种类、洗脱剂配比、上样液流速以及上样量这些单因素对叶酸分离效果的影响的基础上,通过响应面分析找出最优工艺参数。结果表明:产朊假丝酵母Y2.12发酵液分离纯化得到叶酸的最佳工艺条件为:上样液流速为1.26mL/min,发酵液上样量为5.29mL,洗脱剂乙醇与氨水之比1.84:1,所提取的叶酸质量为(101.12±0.13)μg。 Fermented broth of Candida utilis Y2.12 was separated and purified by silica gel column chromatography.The elution effects with different kinds of eluents,proportion of two eluents,flow rate and loading amount were studied.Based on single factor experiments,the proportion of ethanol and ammonia,flow rate,loading amount were analyzed by response surface methodology.The results showed that the optimal separation technological parameters were as follows:1.26mL/min flow rate,5.29mL loading amount,1.84:1 the proportion of ethanol and ammonia.The weight of extracted folic acid was(101.12±0.13)μg.
