摘要
目的:探讨溶血性贫血(溶贫)和再生障碍性贫血(再障)发病过程中红细胞(RBC)膜的构架蛋白与细胞寿命的关系。方法:制备杂种家兔贫血动物模型,用冷冻蚀刻电镜技术和形态计量法观察分析溶贫兔、再障兔和正常兔RBC膜蛋白颗粒的形态、分布、数量和大小。并用^(51)Cr标记技术检测RBC寿命。结果:溶贫兔和再障兔RBC质膜内片劈裂面和外片壁裂面均见蛋白颗粒呈缗钱状、丛簇状分布,数量较正常兔减少(P<0.01);内片劈裂面的颗粒大小和间隙与正常RBC亦有显著性差异(P<0.01)。溶贫组和再障组的RBC寿命明显短于正常组,溶贫组最短。结论:RBC的寿命和功能与其胰蛋白颗粒分布、数量和质量可能有直接的关系。
To explore the relationship between the life span and the structural proteins of red blood cell(RBC) membrane in the rabbits suffering from hemolytic anemia(HA) and aplastic anemia(AA). Methods: The morphology and distribution of the erythrocyte membrane protein granules were observed by freeze fracture etching and transmission electron microscope in the healthy rabbits and the rabbits with HA, AA. The total number and the size of erythrocyte membrane protein granules were measured by morphometry. The 51Cr-RBC method was used to determine the erythrocyte life span of 20 rabbits in each group respectively. Results: The erythrocyte membrane protein granules with HA and AA rabbits were distributed on a string of beads, plexus and groups in protoplasmic face(PF), and extracellular face(EF). The granule numbers from HA and AA were considerably less than that from healthy rabbits (PRO. of). There was a highly significant difference to normal rabbits in the size and space of the PF(P<0. of). The life span of erythrocytes in HA and AA rabbits was shorter than the normal, but the lift span of erythrocytes in HA rabbits was the shortest. Conclusion: There was certain relationship in quantity, quality and distribution of RBC membrane proteins with the lfie span and function of erythrocytes.
