摘要
目的研究竹节参不同提取部位预处理对冠脉结扎诱导大鼠急性心肌缺血损伤的保护作用。方法将50只实验大鼠按完全随机设计分组法分为假手术组、模型组和竹节参不同提取部位组;竹节参不同提取部位组大鼠分别灌胃给予竹节参不同提取部位(乙酸乙酯部位、正丁醇部位和水提部位),给药7 d后制作大鼠急性心肌缺血模型,术后12 h记录大鼠心电图,取血,分离血清后测谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)活性和丙二醛(MDA)、心肌肌钙蛋白I(cTnI)含量,取心脏组织,分别进行心肌组织2,3,5-氯化三苯基四氮唑染色和HE染色。结果竹节参的乙酸乙酯部位组、正丁醇部位组和水提部位均有对抗ST段升高作用,对心肌缺血大鼠心肌梗死面积有较强的降低作用,依次为(10.7±2.4)%、(9.8±2.6)%、(11.0±2.0)%vs(14.8±3.5)%,均具有统计学差异(P<0.05),心肌炎性浸润和溶解坏死现象均有所减轻;在抗氧化损伤和降低cTnI方面,它们能明显增加SOD、GSH-Px酶活性,降低MDA和cTnI含量,与模型组比较,竹节参正丁醇部位均具有统计学差异(P<0.01),水提部位除MDA外,均具有统计学差异(P<0.05,P<0.01)。结论竹节参不同提取部位预处理对冠脉结扎诱导大鼠急性心肌缺血损伤具有较强的保护作用,其中以正丁醇部位最为显著,水提部位次之。
Objective To investigate the protective effects of preconditioning with different fractions extracted from Panax japonicus C.A.Mey on acute myocardial ischemia in rats induced by ligating of the left anterior descending branch(LAD).Methods Fifty Wistar rats were evenly and randomly divided into a sham group,a model group and three treatment groups.The treatment groups were given different fractions extracted from Panax japonicus C.A.Mey,i.g.,an ethyl acetate fraction,an n-butyl alcohol fraction and a water fraction,separately.After preconditioning for 7 d,the acute myocardial ischemia models were constructed by LAD.The electrocardiogram,infarct size and histomorphology were assessed 12 h after LAD.The activities of superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px) as well as the levels of malondialdehyde(MDA) and cardiac troponin I(cTnI) in serum were also measured.Results The ethyl acetate,n-butyl alcohol and water fractions could significantly decrease the ST-segment elevation of electrocardiogram,and remarkably reduce the infarct size [(10.7±2.4)%,(9.8±2.6)% and(11.0±2.0)% vs(14.8±3.5)%,P0.05].The myocardial inflammatory infiltration and lytic necrosis were alleviated due to the three fractions.Besides,the three fractions could significantly enhance the activities of SOD and GSH-Px,and reduce the levels of MDA and cTnI.The effect of the n-butyl alcohol fraction was significant as compared with the model group(P0.05),and that of the water fraction was also significant as compared with the model group except for in the case of MDA(P0.05,P0.01).Conclusion The preconditioning with different fractions extracted from Panax japonicus C.A.Mey has significant protective effects on acute myocardial ischemia in rats.The n-butyl alcohol fraction has the most significant protective effect,followed by the water fraction.
