摘要
目的改进心肌缺血再灌注模型的制备方法,确定判断模型成功的综合指标,为研究心肌缺血再灌注提供更为准确的模型。方法制备家兔心肌缺血再灌注模型,钝性开胸,结扎冠状动脉,45 min后剪断手术线,实现再通,在此期间观测并记录心电图改变,3 h后颈总动脉穿刺取血,分离血清测CK、CK-MB,摘取心脏做常规病理切片。结果结扎后心电图ST段明显抬高,复灌后ST段逐渐回落,血清CK模型组较空白组明显增高,有统计学意义(P<0.05),假手术组变化不明显,CK-MB增高,但无统计学意义(P>0.05),病理切片模型组心肌损伤明显。结论该方法能有效建立心肌缺血再灌注模型,通过指标综合分析,能准确地对模型做出评估。
Objective To improve the method of establishing myocardial ischemia reperfusion injury model,determine the composite indicator to judge model of success,to provide more accurate model for study.Methods The model of myocardial ischemia reperfusion injury in rabbits were established,the chest inactive was opened,the coronary artery was ligated,and the surgery line was cut after 45 min to achieve recanalization.The ECG changes were observed and recorded during this period,neck artery was punctured to get blood after 3 h,serum was separated to measure CK,CK-MB,the heart was procured to make pathological slice.Results The ST segment obviously elevated after being ligated and declined gradually after reperfusion.Model group of serum CK was significantly higher than the blank group,Sham-operated group did not change obviously,CK-MB increased without significance,myocardial injury of the model group was significant in pathological slice.Conclusion The myocardial ischemia reperfusion injury model can be established effectively through this method,an accurate assessment of the model can be made through the comprehensive analysis of indicators.
