摘要
以黄瓜品种津优38中插入缺失(InDel)标记为研究对象,筛选获得能够区分津优38父本、母本及其杂交种的[TT]缺失位点;应用焦磷酸测序技术检测该位点,初步建立基于InDel标记的津优38种子纯度鉴定方法并检测8批种子样品。结果表明,应用焦磷酸测序技术检测[TT]缺失位点能明显区分出津优38父本、母本及杂交种;基于InDel标记的津优38种子纯度鉴定方法具有高准确性、高通量、操作简单、易于自动化等优点;8批种子样品平均种子纯度为97.08%,与SSR鉴定结果(95.83%)基本相符。
An insertion-deletion length polymorphism (InDel-based)marker designed to distinguish Jinyou38 and its parent material had been analyzed by pyrosequencing. The technique of InDel-based pyrosequeneing was applied to assay the seed purity of Jinyou 38 cucumber and the method was used to complete 8 batches hybrid cucumbers. The results showed that parent material and the hybrid cucumber were separated from each other by Indel-based pyrosequencing. The method of InDel-based molecular assay for hybrid cucumber seed purity identification was superior to other methods such as accuracy, high throughout, simple operation, and robotization. The average purity of 8 batches was 97.08 %, which was consistent to the SSR purity.
