摘要
【目的】采用ISSR分子标记对苦瓜杂交一代品种秀玉1号纯度进行鉴定,为生产上提高苦瓜新品种纯度鉴定效率提供参考。【方法】利用ISSR技术和91条引物,对苦瓜品种秀玉1号及其亲本的DNA指纹进行分析。【结果】从91条引物中筛选出两条特征引物ISSR-845和ISSR-891,能同时扩增出F1代及其父母本的多态性条带510和300 bp。其中,ISSR-845能够区分父本机械性混杂,ISSR-891能够准确区分母本自交系与杂种F1代;经群体验证,该标记与田间鉴定结果高度一致。【结论】ISSR技术具有准确、简便、高效、实用性强等优点,可用于生产上苦瓜种子纯度的快速鉴定。
【Objective】In order to improve the efficiency of seed purity identification, seed purity of bitter gourd hybrid Xiuyu 1 was determined using ISSR markers. 【Method】The DNA fingerprints of Xiuyu 1 bitter gourd and its parents were analyzed using 91 primers and ISSR technology. 【Result】The results showed that two primers ISSR-845 and ISSR-891 at 510 bp site and 300 bp site, respectively, were screened out from 91 primers, which could amplify the characteristic polymorphic bands of F1 generation and its parents. ISSR-845 could distinguish the male parent, and ISSR-891 could distinguish the female parent inbred line from Xiuyu 1. Via group verification, the specific markers coincided with those of field identification by morphological detection. 【Conclusion】ISSR technology for seed purity identification of Xiuyu 1 was more accurate, simpler, easier and more practical,and could be used to rapidly and efficiently identify seed purity of bitter gourd in production.
