摘要
目的探讨阿伐他汀对糖尿病大鼠肾脏骨桥蛋白(OPN)表达的影响及其调控的可能机制。方法 45只SD大鼠,随机分为4组:正常组、高脂对照组、糖尿病组和阿伐他汀组。除正常组外,其余3组均用高脂饲料喂养。8周后,糖尿病组与阿伐他汀组腹腔注射链尿佐菌素(STZ)35mg/kg;阿伐他汀组注射STZ 1周后,灌胃给予20 mg/(kg.d)阿伐他汀8周,心脏取血并获肾脏组织。酶法分析血糖、甘油三酯、胆固醇水平。HE染色观察肾脏的病理学变化,PV法免疫组化观察OPN在肾脏中的表达及定位,Westernblot法检测新鲜组织中OPN和pp38表达。结果阿伐他汀可明显降低伴有高脂血症的糖尿病大鼠血清胆固醇浓度,HE、免疫组化和Western blot结果表明:与糖尿病组相比,阿伐他汀可明显减轻肾小管和肾小球的病理损伤程度,下调OPN的表达和p38磷酸化水平,差异有统计学意义(P<0.05)。结论阿伐他汀可能通过降低p38的磷酸化而下调OPN表达。
Objective To investigate the effects of atorvastatin on expression of osteopontin (OPN) in diabetic rats kidney and its regulation mechanism. Methods 45 SD rats were randomly divided into normal group, higt fat group, diabetic group and atorvastatin group. The diabetic group and the atorvastatin group rats were treated with streptozotocin by intraperitoneal injection(35 mg/kg) after they were given 8-week's high fat diet. After a week, the atorvastatin group rats were give atorvastatin 20 rag/( kg · d). The blood and kidney were obtained after 8 weeks. Serum glucose,triglyceride, cholesterol were analyzed. The renal pathology injury was observed by HE staining. The expression of OPN and pp38 were detected by Western blot and immunohistochemical methods. Results Atorvastatin markedly decreased cholesterol and kidney pathology damage. HE, immunohistochemistry and Western blot results showed that the reduction of expression of OPN and pp38 was significant compared with the diabetic group(P 〈 0. 05). Conclusion Atorvastatin maybe decrease the expression of OPN via p38/MAPK pathway.
