茉莉酸甲酯对紫杉醇生物合成的诱导作用 被引量:16

摘要 本文采用分裂素自养型中国红豆杉细胞株 ,研究了在细胞悬浮培养过程中茉莉酸甲酯 ( MJ)对紫杉醇生物合成的诱导作用。结果表明 ,以乙醇为 MJ助溶剂时 ,MJ的诱导作用以剂量为 2 0 0 μmol/L于继代培养开始时加入为最佳 ,此时紫杉醇产量较对照组提高 71.2 %。以吐温为 MJ助溶剂时 ,MJ的诱导作用以剂量为 10μmol/L于继代培养 d2 0加入为最佳 ,此时紫杉醇产量较对照组提高 2 80 .7%。此外 ,本文对 MJ的诱导作用机理进行了探讨。 Taxol is currently the best known drug approved for use in the treatment of ovarian and breast cancer,it also has potential antitumor activities against a variety of solid tumors.However,the supply of the drug from nature resources is limited because Taxus trees grow very slowly and contain only a small amount of taxol.At present, Taxus cell culture has been extensively considered to be one of the most promising methods for obtaining taxol.Methyl jasmonate(MJ)is a kind of plant hormone,several peports have been published indicating that MJ is a signal transducer in plant cell,can induce secondary metaboilte biosynthesis.In this paper,the effect of MJ on taxol biosynthesis was investigated in detail.The results show that MJ can promote the production of taxol in Taxus chinensis cell culture strongly.When MJ was added in the beginning of subculture at concentration of 200 μmol.L -1 with ethanol auxiliary agent,the taxol yield increases by 71 2% compared with control.The best induction effect was obtained when MJ was added on day 20 of subculture at concentration of 10 μmol.L -1 with Tween 80 auxiliary agent,the taxol production increased by 280 7% compared with control.Moreover,the induction mechanism of MJ was also analyzed.It is considered that the induction mechanism of MJ is different from that of fungi elicitors,that is,both MJ and fungi elicitors can induce taxol biosynthesis,but the inhibition of fungi elicitors on cell growth is far stronger than that of MJ.Furthermore,the concentration of fungi elicitors is inversely related to the cell growth,while the concentration of MJ isn't correlative with the cell growth,and the inhibition of MJ on Taxus cell growth can be reversed by cytokinin.The paper suggests that MJ play a role of signal transducer in disease resistance reaction induced by fungi elicitors.
